The Leica MZ16FA Fluorescence Stereo Microscope/Macroscope

Powering Up the System:

• Note that the two microscope switches and the b/w camera must be on before the software is started
• Leica EL6000 metal halide lamp
  • Turn on green power switch
  • Ensure shutter switch is pushed in.  Yellow shitter open light should be on
• MZ61FA
(microscope stand)
• Turn on switch at back of base on right side
• Turn on switch on top of focus drive
• Ensure computer is on
• Ensure monitor is on
  • Ensure video cable on back of monitor is from the computer and not the VCR
  • If you use the VCR please put the computer's video connector back when your done
• Turn on Hamamatsu OrcaER camera
  • Ensure the Hamamatsu OrcaER is the camera on the top left c-mount port on the scope body.
  • If not -
    • loosen locking screw
    • pull out little video camera and c-mount tube
    • unscrew c-mount
    • unscrew cap that should be on OrcaER
    • screw cap into small video camera
    • attach c-mount to OrcaER
    • insert OrcaER and c-mount into top left port on microscope body
    • tighten locking screw
• Turn off or on incident light lamp - KL1500
• Log in to computer using your user name, password and the MHmicroscopy domain
• Start C-Imaging
• Turn off transmitted light by pushing button on the rear of the right side of the base
• Change filters on scope body if necessary & run scope hardware setup on tab in acquisition window of c-imaging

Taking Pictures (real brief notes)

• Select camera
• Select channels and filters
• Go live
• Set exposures and gains for each channel
• If transmitted light - do a white balance on a white/blank

Powering Down the System:

• Exit C-Imaging (don't forget to save any pictures)
• Power off at top of focus drive
• Power off scope body switch at rear right side of base
• Turn off Leica EL600 metal halide lamp
• Please sign log book
